Jacob Kane was a male human born on Earth as a member of the Kane family.
Post-Flashpoint
Following the Flashpoint, a new version of reality was created with a different history of events. Jacob Kane was noted to had been the brother of Phillip Kane and him having a sister in Martha Kane. (Detective Comics v1 #939)
After being killed in Crime Alley, Jacob was noted to had attended his sister Martha's funeral alongside his family. (Detective Comics v1 #939)

- In Batwoman, Jacob Kane appeared in the live-action television series set in the Arrowverse where he was portrayed by actor Dougray Scott. He was the father of Kate and Beth Kane where he came to head a private military firm called Crows Security. Jacob was born in Gotham City into the Kane family and had an older sister, Martha. In his youth, Jacob became an uncle when Martha married Thomas Wayne and had a son, Bruce Wayne. Sometime afterwards, his sister and brother-in-law were both killed in a mugging. By the time he was an adult, Jacob had joined the United States Army as a field agent in the 3rd Special Forces Group, earning a decorated career and eventually rising through the USSOCOM ranks to become a colonel. After his service, Jacob fell in love with and married Gabi Goldstein. On January 26, 1990, they had twin daughters, Kate and Beth, short for Katherine and Elizabeth respectively, whom he loved deeply. On January 26, 2003, Jacob and Gabi held a Bat Mitzvah for their daughters at their synagogue to celebrate Kate and Beth's 13th birthday. Jacob's car was filled with so many birthday gifts that there wasn't enough room left for passengers. He joked about having to give some of the presents away, to which Kate and Beth assured him they could go home with Gabi. Musing how the girls were now "officially young women", Jacob kissed his daughters and wife goodbye before driving off.

- In Batman: Bad Blood, Jacob Kane appeared in the setting of the animated film where he was voiced by actor Geoff Pierson. When his wife Gabrielle, daughters Katherine and Elizabeth Kane were kidnapped on the twins' 12th birthday and held for ransom. Colonel Kane led the rescue team. In the attempt, Gabrielle and Elizabeth Kane were killed. Colonel Kane shot the men who were about to kill Kate. Colonel Kane checked on Katherine. Some time later, Katherine decided to follow in her father's foot steps and enrolled at West Point but was ultimately expelled. He later learned of her homosexuality and accepted it. Kane knows of Katherine's costumed career as Batwoman and helps her gather intelligence from time to time using his old contacts.

- In Gotham Knights, Jacob Kane appeared in the setting of the video game where he was voiced by actor Tommie Earl Jenkins. Col. Jacob Kane was a former soldier of the United States Army and CEO of Kane Industries. He was also married to Catherine Kane, who was the new Commissioner of the GCPD. Unknown to anyone, he was a member of the Court of Owls where whilst wearing a mask he assumed the position of Voice of the Court. Upon discovery of his affiliation with the Court of Owls however, the Knights looked to prosecute him, since they had evidence to prove his involvement in the death of Dr. Kirk Langstrom and his assistant. They obtained an arrest warrant from Det. Renee Montoya and signed by judge Elena Moreno. With the warrant and the resources to deal a crippling blow to Kane, they infiltrated Kane Industries' headquarters and arrested him. He was escorted out of his building by Montoya and scolded by his wife, for his involvement in murder. Before Kane could enter a courtroom however, Talia al Ghul, the daughter of the late Ra's al Ghul, killed him with an arrow to the chest.
